In other words, which gift has the most chance of getting me the Endless Tonic?

Only the Giant Gift has a chance to drop the tonic.  Otherwise, you need to craft it with the Toy Frames.  If you craft it, you won't be able to craft the mini-pet.

Darn I used up one of my toy frames already. Is all hope lost for me for crafting it?

Since the completion of the dungeon is account-bound, and the frame itself is also account-bound: yes, you won't be able to get it anymore.

I don't know if the resulting tonic is account-bound as well, but I assume it is.

There is always the "Toypocalypse" event, which might or might not award extra frames. We'll know when Trixx lands in LA...
