Repair your GW2 installation. Make a shortcut to Gw2.exe, edit the "Target" and add -repair. Should look something like this (path to gw2 may vary):
Target: "C.\Program Files\Guild Wars 2\Gw2.exe" -repair

Very similar error for me:

*--> Crash <--*
Assertion: Failed to load file.
File: ..\..\..\Services\LoadHandle\LoadHandle.cpp(144)

But even after repair it is not working correctly. Now, I can either run the repair mode, it crashes before starting checking file integrity! Any suggestion? Should I uninstall and reinstall the game?

You could try -image instead of -repair.

And of course there's the usual stuff...
- clean the dust out of the machine and check that all fans are working.
- make sure all your drivers are up to date
- make sure you have enough free space on your drive.
- check for loose cables, re-seat the RAM and video card

what does the -image option do?
My PC has less than 2 months ;) I hope it is not already some hardware problem :)

"-image" makes sure that the entire, updated, game is installed. It basically makes sure that your entire installation is up to date. It can take a long time.

I reinstalled GW2 from scratch yesterday and it did not work. Temperature are under control, speccy does not see anything strange. A part from the SSD at 128 degrees, but I believe that is a mistake, because everything works fine (and the case is not on fire also).

Try this: create a .bat file with the following lines in it
@ECHO OFF
cmd.exe /c start "Run with Dual-Core" /affinity 3 "Gw2.exe"
And save the .bat to the same folder that the client is in. and then use that to launch gw2.

Found it somewhere and it did not stop the crashes entirely, but it did make it less frequent to happen.
